DESCRIPTION
What It Does: Provides nuclease-resistant antisense gene silencing targeting specific pro-inflammatory or pro-aging mrna sequences โ the phosphorothioate modification extending in-skin half-life from minutes (unmodified dna) to hours for effective gene knockdown.
Why It's Used: Phosphorothioate modification is the foundational chemistry of all approved antisense drugs โ the sulfur replacement converting rapidly degraded dna to a nuclease-resistant therapeutic oligonucleotide applicable to cosmetic gene targeting.
How It Works: Phosphorothioate (ps): phosphate p=o โ p=s replacement โ sulfur's larger ionic radius and lower electronegativity โ reduced rnase h binding but maintained watson-crick hybridisation โ rnase h-mediated mrna cleavage at dna/rna hybrid. nuclease resistance: ps sulfur doesn't coordinate znยฒโบ/mgยฒโบ in nuclease active sites โ >100ร slower degradation versus unmodified phosphodiester.
