DESCRIPTION
What It Does: Provides sglt-mediated glucose transport reduction for anti-glycation protection of skin collagen, tyrosinase inhibitory brightening from the dihydrochalcone pharmacophore, and potent antioxidant radical scavenging.
Why It's Used: Phloridzin is historically significant as the first compound to demonstrate sglt inhibition (1835) โ the same mechanism later developed into the gliflozin pharmaceutical class. topically, the sglt inhibition reduces keratinocyte glucose uptake reducing local glycation of sc proteins.
How It Works: Phloridzin: competitive sglt1/2 inhibition (kd ~200 nm) โ reduced intracellular glucose โ reduced maillard reaction on skin proteins โ anti-glycation. phloretin aglycone (from skin glucosidase hydrolysis): competitive tyrosinase inhibition (ic50 ~30 ฮผm) โ brightening. dihydrochalcone alpha,beta-unsaturated carbonyl: radical scavenging from phenol h-atom donation.
