What It Does: Provides intense trpm8-mediated cooling sensation at concentrations 10-100ร lower than menthol โ the carboxamide pharmacophore providing high receptor affinity with minimal peppermint fragrance.
Why It's Used: Synthetic cooling agents separate cooling function from menthol fragrance โ enabling cooling product positioning in applications where maximum cooling with minimal peppermint scent is desired.
How It Works: Cyclohexane/menthane carboxamide scaffold binds trpm8 hydrophobic pocket (s1-s4 transmembrane domain): carboxamide c=o h-bonds tyr745; alkyl chain occupies lipophilic groove. higher binding affinity than menthol (ec50 0.1-1 ฮผm vs menthol 10 ฮผm) โ equivalent cooling at 10-100ร lower concentration.
